Airborne laser reveals city under Cambodian earth

SYDNEY (AP) ? Airborne laser technology has uncovered a network of roadways and canals, illustrating a bustling ancient city linking Cambodia's famed Angkor Wat temples complex.

The discovery was announced late Monday in a peer-reviewed paper released early by the journal Proceedings of the National Academy of Sciences. The laser scanning revealed a previously undocumented formal urban planned landscape integrating the 1,200-year-old temples.

The airborne lasers produced a detailed map of a vast cityscape, including highways and previously undiscovered temples, hidden beneath dense vegetation atop Phnom Kulen mountain in Siem Reap province. It was the lost city of Mahendraparvata.

"What we have now with this instrument is just 'bang' ? all of a sudden, an immediate picture of an entire city that people didn't know was there before, which is remarkable," University of Sydney archaeologist Damian Evans, the study's lead author, told Australia's The Age in a video interview from Cambodia. "So instead of this kind of very long gradual process, you have this kind of sudden eureka moment where you bring the data up on screen the first time and there it is ? this ancient city very clearly in front of you."

The laser technology, known as lidar, works by firing laser pulses from an aircraft to the ground and measuring the distance to create a detailed, three-dimensional map of the area. It's a useful tool for archaeologists because the lasers can penetrate dense vegetation and cover swaths of ground far faster than they could be analyzed on foot. Lidar has been used to explore other archaeological sites, such as Stonehenge.

In April 2012, the Australian researchers loaded the equipment onto a helicopter, which spent days crisscrossing the dense forests from 800 meters above the ground. The team then confirmed the findings with an on-foot expedition through the jungle.

"We had reasonable expectations, I guess, of what we would find using the lidar data, but what we've ended up with has just blown our minds," Evans told The Age. "It's just absolutely incredible what we can see."

The researchers theorize the civilization at Mahendraparvata eventually collapsed because of deforestation and broken canals and reservoirs.

'World War Z' draws massive crowd in NYC

NEW YORK (AP) ? Thousands of screaming fans brought Times Square to a standstill as they waited to catch a glimpse of Brad Pitt at the premiere of his new thriller, "World War Z."

The 49-year old actor took his time to talk to reporters on the appropriately-designed "Z-shaped" red carpet, often breaking away to sign autographs and pose for photographs with fans.

"We're not doing this small... we're going big this summer," Pitt said of the premiere that resembled a music festival.

Pitt stars in and produced the apocalyptic thriller based on the book by Max Brooks. He's the son of the legendary Mel Brooks.

Pitt plays a former United Nations investigator who traverses the world in a race against time to stop a zombie pandemic that is threatening to decimate humanity.

"We liked this idea of taking a genre and using that as, I guess, a metaphor for pandemics, and what if one of these pandemics jumped the tracks. Would we be ready? What countries would be in better shape, what countries would take the biggest hit," Pitt said of the zombie thriller.

While Pitt sees the film as more of a summer thriller than a cautionary tale, he's nonetheless intrigued by the political subtext.

"A movie is only good if it speaks about our time, if it's personal in some way, and it plugs into the zeitgeist to irritate a little bit. I think we got that here," he said.

As for letting his children see it, Pitt feels the movie is not appropriate for the younger ones.

"My two oldest I've let see it, and they've loved it. Parental guidance, it's called parental guidance," Pitt said of only allowing Maddox, 11, and Pax, 8, to see the film.

Pitt, and fiance' Angelina Jolie, have six children.

Jolie has long been recognized for her humanitarian work, and served as Goodwill Ambassador for the United Nations High Commissioner for Refugees, or UNHCR. In 2012, she was promoted to Special Envoy.

So how does Jolie feel about Pitt representing the U.N in the film?

Pitt laughed, saying: "She just didn't want me to embarrass them."

Poll: Hong Kongers would not back extradition of Edward Snowden

About 50 percent say the NSA whistleblower should not be surrendered, 17.6 percent said he should be turned over, and a third aren't sure yet, according to poll published today.

By a three-to-one margin, Hong Kongers do not want their government to hand over NSA whistleblower Edward Snowden if Washington demands his extradition.

Whether it is because they support Mr. Snowden?s free-speech and privacy agendas, or because they are upset by his claims that the US National Security Agency has been hacking into Hong Kong?s computer network, 49.9 percent of people asked in a poll published here?Sunday?said he should not be surrendered. Another 17.6 percent said he should be turned over. A third of respondents had not made up their minds.

?Nobody welcomes a fugitive, but now he is here we have to safeguard his rights,? said Freddy Chu, a young privacy activist, as he brandished a photo of Snowden at a small rally in support of the American in central Hong Kong?on Saturday.

Snowden is believed to be in hiding somewhere in Hong Kong, from where he divulged his identity to The Guardian newspaper a week ago. Since then, in an interview with the South China Morning Post, he has accused the NSA of hacking into the backbone of Hong Kong?s Internet system.?

?He is welcome to Hong Kong,? said another demonstrator, James Hon, as he helped hold up a banner belonging to the League in Defense of Hong Kong?s Freedoms. ?He is upholding our core values ? freedom of expression and privacy. He is a brother.??

Snowden?s presence here puts the former British colony in a difficult spot, potentially subject to pressure from both Washington and Beijing. For the time being, the United States has not lodged an extradition request and Chinese officials have not tipped their hand about what they think should happen to Snowden. But many Hong Kongers are uncomfortable.?

That may explain the low turnout at Saturday?s demonstration outside the US Consulate. Persistent rain did not help, but few people here see Snowden?s fate as very important to their own lives.?

Still, his suggestions that the NSA has been hacking in Hong Kong have won him a measure of sympathy. ?When we learned that they had hacked into our Internet hub at the Chinese University of Hong Kong, all of us with a computer felt we had been targeted,? said Yves Azemar, a French expatriate dealer in rare books.?

Snowden?s presence here also draws international attention to Hong Kong?s unique status as a ?special administrative region? of China, where the rules are very different from the mainland.?

?This is a golden opportunity for Hong Kong to explain to the world ? that we still enjoy judicial autonomy,? says Alan Leong, a legislator and head of the pro-democracy Civic Party. ?It?s a chance to say how proud we are that Snowden chose Hong Kong as a refuge.??

ENNISKILLEN, Northern Ireland (AP) ? President Barack Obama declared peace in Northern Ireland a "blueprint" for those living amid conflict around the world, while acknowledging that the calm between Catholics and Protestants will face further tests. Summoning young people to take responsibility for their country's future, Obama warned there is "more to lose now than there's ever been."

"The terms of peace may be negotiated by political leaders, but the fate of peace is up to each of us," Obama said Monday during remarks at Belfast's Waterfront Hall. The glass-fronted building would never have been built during the city's long era of car bombs.

Obama arrived in Northern Ireland Monday morning after an overnight flight from Washington. Following his speech to about 1,800 students and adults, he flew to a lakeside golf resort near Enniskillen, passing over a sweeping patchwork of tree-lined farms as he prepared to meet with other leaders of the Group of 8 industrial nations on Syria, trade and counterterrorism. One-on-one meetings with Russian President Vladimir Putin, British Prime Minister David Cameron and Italian Prime Minister Enrico Letta were all on the agenda for Monday.

Cameron selected Enniskillen as the site of this year's meeting as a way to highlight Northern Ireland's ability to leave behind a four-decade conflict that claimed 3,700 lives.

Significant progress has been made in the 15 years since the U.S.-brokered Good Friday Accords, including a Catholic-Protestant government and the disarmament of the IRA and outlawed Protestant groups responsible for most of the 3,700 death toll. But tearing down Belfast's nearly 100 "peace lines" ? barricades of brick, steel and barbed wire that divide neighborhoods, roads and even one Belfast playground ? is still seen by many as too dangerous. Obama cited that playground in his speech, lauding an activist whose work led to the opening of a pedestrian gate in the fence.

Acknowledging the reality of a sometimes-fragile peace, Obama recalled the Omagh bombings that killed 29 people and injured hundreds more. It was the deadliest attack of the entire conflict and occurred after the Good Friday deal.

Peace will be tested again, Obama said in Belfast.

"Whenever your peace is attacked, you will have to choose whether to respond with the same bravery that you've summoned so far or whether you succumb to the worst instincts, those impulses that kept this great land divided for too long. You'll have to choose whether to keep going forward, not backward," he said.

Last month, the Catholic and Protestant leaders of Northern Ireland's unity government announced a bold but detail-free plan to dismantle all peace lines by 2023. British Prime Minister David Cameron formally backed the goal Friday, and Obama followed with his own endorsement Monday.

The president specifically endorsed an end to segregated housing and schools, calling it an essential element of lasting peace.

"If towns remain divided ? if Catholics have their schools and buildings, and Protestants have theirs, if we can't see ourselves in one another, if fear or resentment are allowed to harden ? that too encourages division. It discourages cooperation," Obama said.

One symbol of that effort to end the segregation was on display as Obama spoke to an audience that brought together students from both faiths, effectively integrating Northern Ireland's schoolchildren if just for a morning.

Drawing on America's own imperfect battle with segregation, Obama recalled how a century after the U.S. Civil War, the nation he leads is still not fully united. His own parents ? a white woman from Kansas and a black man from Kenya ? would not have been able to marry in some states, Obama said, and he would have had a hard time casting a ballot, let alone running for office.

"But over time, laws changed, and hearts and minds changed, sometimes driven sometimes by courageous lawmakers, but more often driven by committed citizens," he said.

Though Obama did not specifically mention Syria, his remarks on Northern Ireland recalled the fierce conflict there that has so far resulted in 93,000 deaths. For those looking for a way out of conflict, Obama said Northern Ireland is "proof of what is possible."

Obama and other G-8 leaders were expected to discuss Syria Monday night over a working dinner. Obama will be looking to Britain and France to join him in sending weapons to the Syrian opposition.

Casting a shadow over the summit are new revelations by the Guardian newspaper that the British eavesdropping agency GCHQ repeatedly hacked into foreign diplomats' phones and emails when the U.K. hosted international conferences, including a 2009 Group of 20 summit in London. The report follows recent disclosures about the U.S. government's own surveillance programs and could lead to awkward conversation as the leaders open another international gathering that Britain is hosting.

Despite an agenda devoted to trade, economic growth and international tax issues, the G-8 will be eclipsed by discussions over how to address the two-year-old civil war in Syria and the decision by the United States to begin supplying rebels with military aid.

Obama's meeting with Russia's Putin later Monday will highlight the rift between their countries in addressing fierce fighting in Syria. While Putin has called for negotiated peace talks, he has not called for Syrian President Bashar Assad to leave power, and he remains one of Assad's strongest political and military allies.

In a likely preview of his discussions with Obama, Putin defended Russia's continuing supply of weapons to Assad's military in a meeting Sunday with Cameron, the British leader.

Putin said Russia was providing arms "to the legitimate government of Syria in full conformity with the norms of international law."

The White House is not expecting any breakthrough with Putin on Syria during Putin's meeting with Obama.

Obama is making his first visit to Northern Ireland, though he visited the neighboring Republic of Ireland in 2011. That trip included a public speech in the center of Dublin, as well as a stop in the village of Moneygall, where Obama's great-great-great grandfather was born. The president called that visit "magical."

First lady Michelle Obama and daughters Malia and Sasha, who also made the trip from Washington, were to spend Monday and Tuesday in Dublin while the president attended the G-8 summit. Later Tuesday, the first family departs for Germany, where the president will meet with Chancellor Angela Merkel and speak at the Brandenburg Gate.

'Undruggable' cancer may be druggable after all: New target identified

June 17, 2013 ? Harvard Stem Cell Institute (HSCI) researchers have identified in the most aggressive forms of cancer a gene known to regulate embryonic stem cell self-renewal, beginning a creative search for a drug that can block its activity.

The gene, SALL4, gives stem cells their ability to continue dividing as stem cells rather than becoming mature cells. Typically, cells only express SALL4 during embryonic development, but the gene is re-expressed in nearly all cases of acute myeloid leukemia and 10 to 30 percent of liver, lung, gastric, ovarian, endometrial, and breast cancers, strongly suggesting it plays a role in tumor formation.

In work published in the New England Journal of Medicine, two HSCI-affiliated labs -- one in Singapore and the other in Boston -- show that knocking out the SALL4 gene in mouse liver tumors, or interfering with the activity of its protein product with a small inhibitor, treats the cancer.

"Our paper is about liver cancer, but it is likely true about lung cancer, breast cancer, ovarian cancer, many, many cancers," said HSCI Blood Diseases Program leader Daniel Tenen, who also heads a laboratory at the Cancer Science Institute of Singapore (CSI Singapore). "SALL4 is a marker, so if we had a small molecule drug blocking SALL4 function, we could also predict which patients would be responsive."

Studying the therapeutic potential of a transcription factor is unusual in the field of cancer research. Transcription factors are typically avoided because of the difficulty of developing drugs that safely interfere with genetic targets. Most cancer researchers focus their attention on kinases.

The HSCI researchers' inquiry into the basic biology of the SALL4 gene, however, revealed another way to interfere with its activity in cancer cells. The gene's protein product is responsible for turning off a tumor-suppressor gene, causing the cell to divide uncontrollably. Using this knowledge, the researchers demonstrated that targeting the SALL4 protein with druglike molecules could halt tumor growth. "The pharmaceutical companies decided that if it is not a kinase and it is not a cell surface molecule, then it is 'undruggable,' " Tenen said. "To me, if you say anything is 'undoable,' you are limiting yourself as a biomedical scientist."

Earlier this year, Tenen's co-author, HSCI-affiliated faculty member Li Chai, a Harvard Medical School assistant professor of pathology at Brigham and Women's Hospital, published a paper in the journal Blood, reporting that a SALL4 inhibitor has similar treatment potential in leukemia cells.

Chai took blood samples from patients with acute myeloid leukemia, treated the leukemic cells with the inhibitor that interferes with SALL4 protein activity, and then transplanted the blood into mice. The result was a gradual regression of the same cancer in mice.

"I am excited about being on the front line of this new drug development," Chai said. "As a physician-scientist, if I can find a new class of drug that has very low toxicity to normal tissues, my patients can have a better quality of life."

Chai and Tenen are now working with HSCI Executive Committee member Lee Rubin, the Harvard Institute of Chemistry and Cell Biology, and James Bradner of Dana-Farber Cancer Institute, another HSCI-affiliated faculty member, to overcome the technical challenges of drug development and demonstrate the potential of SALL4 interference to treat other forms of cancer.

"I think as academics, we seek to engage drug companies because they can do these types of things better than we can," Tenen said. "But, also as an academic, I want to go after the important biologic targets that are not being sought after by the typical drug company -- because if we do not, who will?"

To ease shortage of organs, grow them in a lab?

NEW YORK (AP) ? By the time 10-year-old Sarah Murnaghan finally got a lung transplant last week, she'd been waiting for months, and her parents had sued to give her a better shot at surgery.

Her cystic fibrosis was threatening her life, and her case spurred a debate on how to allocate donor organs. Lungs and other organs for transplant are scarce.

But what if there were another way? What if you could grow a custom-made organ in a lab?

It sounds incredible. But just a three-hour drive from the Philadelphia hospital where Sarah got her transplant, another little girl is benefiting from just that sort of technology. Two years ago, Angela Irizarry of Lewisburg, Pa., needed a crucial blood vessel. Researchers built her one in a laboratory, using cells from her own bone marrow. Today the 5-year-old sings, dances and dreams of becoming a firefighter ? and a doctor.

Growing lungs and other organs for transplant is still in the future, but scientists are working toward that goal. In North Carolina, a 3-D printer builds prototype kidneys. In several labs, scientists study how to build on the internal scaffolding of hearts, lungs, livers and kidneys of people and pigs to make custom-made implants.

Here's the dream scenario: A patient donates cells, either from a biopsy or maybe just a blood draw. A lab uses them, or cells made from them, to seed onto a scaffold that's shaped like the organ he needs. Then, says Dr. Harald Ott of Massachusetts General Hospital, "we can regenerate an organ that will not be rejected (and can be) grown on demand and transplanted surgically, similar to a donor organ."

That won't happen anytime soon for solid organs like lungs or livers. But as Angela Irizarry's case shows, simpler body parts are already being used as researchers explore the possibilities of the field.

Just a few weeks ago, a girl in Peoria, Ill., got an experimental windpipe that used a synthetic scaffold covered in stem cells from her own bone marrow. More than a dozen patients have had similar operations.

Dozens of people are thriving with experimental bladders made from their own cells, as are more than a dozen who have urethras made from their own bladder tissue. A Swedish girl who got a vein made with her marrow cells to bypass a liver vein blockage in 2011 is still doing well, her surgeon says.

In some cases the idea has even become standard practice. Surgeons can use a patient's own cells, processed in a lab, to repair cartilage in the knee. Burn victims are treated with lab-grown skin.

In 2011, it was Angela Irizarry's turn to wade into the field of tissue engineering.

Angela was born in 2007 with a heart that had only one functional pumping chamber, a potentially lethal condition that leaves the body short of oxygen. Standard treatment involves a series of operations, the last of which implants a blood vessel near the heart to connect a vein to an artery, which effectively rearranges the organ's plumbing.

Yale University surgeons told Angela's parents they could try to create that conduit with bone marrow cells. It had already worked for a series of patients in Japan, but Angela would be the first participant in an American study.

"There was a risk," recalled Angela's mother, Claudia Irizarry. But she and her husband liked the idea that the implant would grow along with Angela, so that it wouldn't have to be replaced later.

So, over 12 hours one day, doctors took bone marrow from Angela and extracted certain cells, seeded them onto a 5-inch-long biodegradable tube, incubated them for two hours, and then implanted the graft into Angela to grow into a blood vessel.

It's been almost two years and Angela is doing well, her mother says. Before the surgery she couldn't run or play without getting tired and turning blue from lack of oxygen, she said. Now, "she is able to have a normal play day."

This seed-and-scaffold approach to creating a body part is not as simple as seeding a lawn. In fact, the researchers in charge of Angela's study had been putting the lab-made blood vessels into people for nearly a decade in Japan before they realized that they were completely wrong in their understanding of what was happening inside the body.

"We'd always assumed we were making blood vessels from the cells we were seeding onto the graft," said Dr. Christopher Breuer, now at Nationwide Children's Hospital in Columbus, Ohio. But then studies in mice showed that in fact, the building blocks were cells that migrated in from other blood vessels. The seeded cells actually died off quickly. "We in essence found out we had done the right thing for the wrong reasons," Breuer said.

Other kinds of implants have also shown that the seeded cells can act as beacons that summon cells from the recipient's body, said William Wagner, director of the McGowan Institute for Regenerative Medicine at the University of Pittsburgh. Sometimes that works out fine, but other times it can lead to scarring or inflammation instead, he said. Controlling what happens when an engineered implant interacts with the body is a key challenge, he said.

So far, the lab-grown parts implanted in people have involved fairly simple structures ? basically sheets, tubes and hollow containers, notes Anthony Atala of Wake Forest University whose lab also has made scaffolds for noses and ears. Solid internal organs like livers, hearts and kidneys are far more complex to make.

His pioneering lab at Wake Forest is using a 3-D printer to make miniature prototype kidneys, some as small as a half dollar, and other structures for research. Instead of depositing ink, the printer puts down a gel-like biodegradable scaffold plus a mixture of cells to build a kidney layer by layer. Atala expects it will take many years before printed organs find their way into patients.

Another organ-building strategy used by Atala and maybe half a dozen other labs starts with an organ, washes its cells off the inert scaffolding that holds cells together, and then plants that scaffolding with new cells.

"It's almost like taking an apartment building, moving everybody out ... and then really trying to repopulate that apartment building with different cells," says Dr. John LaMattina of the University of Maryland School of Medicine. He's using the approach to build livers. It's the repopulating part that's the most challenging, he adds.

One goal of that process is humanizing pig organs for transplant, by replacing their cells with human ones.

"I believe the future is ... a pig matrix covered with your own cells," says Doris Taylor of the Texas Heart Institute in Houston. She reported creating a rudimentary beating rat heart in 2008 with the cell-replacement technique and is now applying it to a variety of organs.

Ott's lab and the Yale lab of Laura Niklason have used the cell-replacement process to make rat lungs that worked temporarily in those rodents. Now they're thinking bigger, working with pig and human lung scaffolds in the lab. A human lung scaffold, Niklason notes, feels like a handful of Jell-O.

Cell replacement has also worked for kidneys. Ott recently reported that lab-made kidneys in rats didn't perform as well as regular kidneys. But, he said, just a "good enough organ" could get somebody off dialysis. He has just started testing the approach with transplants in pigs.

Ott is also working to grow human cells on human and pig heart scaffolds for study in the laboratory.

There are plenty of challenges with this organ-building approach. One is getting the right cells to build the organ. Cells from the patient's own organ might not be available or usable. So Niklason and others are exploring genetic reprogramming so that, say, blood or skin cells could be turned into appropriate cells for organ-growing.

Others look to stem cells from bone marrow or body fat that could be nudged into becoming the right kinds of cells for particular organs. In the near term, organs might instead be built with donor cells stored in a lab, and the organ recipient would still need anti-rejection drugs.

How long until doctors start testing solid organs in people? Ott hopes to see human studies on some lab-grown organ in five to 10 years. Wagner calls that very optimistic and thinks 15 to 20 years is more realistic. Niklason also forecasts two decades for the first human study of a lung that will work long-term.

But LaMattina figures five to 10 years might be about right for human studies of his specialty, the liver.

"I'm an optimist," he adds. "You have to be an optimist in this job."
